Description: To further define whether the miRNAs were present within the mitochondria, we used deoxycholic acid (DCA) to induce opening of the mitochondrial megapore channel33 and induction of the mitochondria permeability transition (MPT) in the isolated mitochondria.This disruption of mitochondrial integrity resulted in a significant loss of miRNAs by RT-PCR in the DCA treated mitochondria relative to the control vehicle group. The release of miR-130a and b, miR-320 and miR-494 was about 50% and that of miR-140 almost 80%.
