Description: miR-501-3p expression is up-regulated locally in dendrites through the NMDAR subunit GluN2A, and this regulation is required for NMDA-induced suppression of GluA1 expression and long-lasting remodeling of dendritic spines. Our further analysis of miR-501-3p shows that it is increased locally in dendrites after NMDAR activation and that this up-regulation of miR-501-3p is required for NMDAR-dependent inhibition of GluA1 expression, long-lasting spine shrinkage, and elimination. These NMDA-induced changes were preserved in the CA1 neuropil of the slice in which cell bodies were removed before NMDA stimulation (Fig. 4, E-G), suggesting that miR-501-3p can repress GluA1 protein expression locally in dendrites.
